See if I have a basic understanding of Share/Folders and a ? about moving Shares

A Share is like a disk mount (Mac) or a virtual HD view (Windows) and can span, occupy by itself and/or share with other Share(s) physical HDs and grow/shrink as files are added/deleted.

A folder will restrict all sub folders and files to the physical HD it is actually located.

I now have 3-400GB HDs for 800GB storage. I moved my iTunes library (about 425GB) to my unRAID under a Share named Music. I also copied about 200GB of HD trailers under a Share named Trailers. Now I'm about to add 2-1TB,1-750GB HD.  My music server is a whole house and runs 24/7.  Right now my Music Share  iTunes library is spread across 2 physical HD's.  After I add the new HDs I would like to move the Music Share to the 750GB HD so only 1 HD needs to spin all the time.  Do I stipulate the HD in the following screen and if so what is the syntax?

Just put in the disk you want to use in the Included Disk(s) spot. For example "disk3".

You will have to manually move the data already on the other disks to the disk you want to use and then delete the directories off of the other disks.
