Yes.

 

I would suggest you print down the the 'Main'--'Array Devices' page on the GUI before you do it as a tiny bit of insurance.  You should not need it. 

 

By the way, why are you moving your drives off of the MB board ports?  Most people populate those first and then add in the cards when the number of drives exceed the available ports.

You may get better performance by shifting only a few of your HD's onto the cards.  Pcie is a complex animal in that you can mix hardware that meets the various versions of the spec but the speed then defaults to the slowest one.  See here for an introduction:
