Since you posted in the Hardware forum, here's the answer pertaining to the hardware aspects.

 

It is up to the manufacturers of the motherboards to ensure they use UEFI instead of BIOS to boot from the drives in addition to using SATA controllers capable of working with such large drives. I have not seen any release statements by any of the big motherboard makers proclaiming ready support for drives larger than 2.1 TB. This is why the existing 3TB drives are either external and/or include their own SATA controller.
