Disk encryption question- encrypt a working drive

I have existing disk, currently not encrypted.  A share uses this disk, and this disk alone.  The share is protected.  I'd like to make sure when this drive give up on me, contents won't be readable.  Do I achieve this using disk encryption?  Can I do this on a drive that is in use?
