Plex with HandBrake converted files

I hope that is a very quick question

 

I have a source file of: with a total disk space size of 7gb 

I would like to reduce this down to 500mb to 1gb, but every time I have done, the file is watchable on my PC but when I move it into a folder on PLEX then plex will not see it.

 

so far I have tried the following defaults -  Very Fast 720p , HQ 720p30 , Matroska h.264 MKV 70p30 

and each time I am getting the same,   I can watch it on my PC but not in Plex.

 

any ideals ?

Update.  After a lot of testing, i ended up rebooting my plex and then server. 

And after do this my plex ended up seeing my videos.

 

Why. No ideal.
